Topically applied B vitamins such as niacinamide has been shown to help skin retain moisture. It also has anti-inflammatory properties to soothe dry and irritated skin. NADH and NADPH are 2 important co- 3enzymes, which contribute to the production of cellular energy and lipids (responsible for cell growth). With a sufficient supply of both enzymes, your skin creates an effective barrier to external factors that influence the aging process. However, the levels of both NADH and NADPH decline as we grow older but when applied topically, vitamin B3 seems to reduce their rate of decline.
This action contributes to the support of the skin’s barrier against pollutants and other irritants, which can result in helping to manage many skin conditions, such as acne, the redness associated with rosacea and other inflammatory signs found on the skin. Vitamin B3 improves skin moisturisation and reduces transepidermal water loss (TEWL) – skin softness, suppleness and skin hydration are related to the barrier properties of the horny layer.
It is known that several lipids such as fatty acids and ceramides are critical for the structural and functional integrity of the stratum corneum. The skin barrier function can be assessed by transepidermal water loss (TEWL) measurements. A study conducted by Tanno et al. showed that 2% niacinamide reduced the TEWL by 24% in 4 weeks. At the same time free fatty acids and ceramides in stratum corneum were boosted by 67% and 34%, respectively.
Also, vitamin B3 has shown to be useful for encouraging the production of natural emollients that soften the skin and help it to stay hydrated. Vitamin B3 prevents skin aging and improves the appearance of aging skin by stimulating new fibroblasts and collagen secretion. Skin aging is characterized by major skin changes like reduced skin elasticity, poorer structure and appearance of wrinkles. An important factor is the gradual loss of collagen and elastin fibers synthesised in fibroblasts. A recommended strategy in preventing skin aging is to reduce collagen breakdown, while increasing fibroblasts.
Studies with human fibroblasts showed that niacinamide stimulates new fibroblasts by 20% and collagen secretion by 54%. A group of 50 women between the ages of 40 and 60 participated in a double-blind, placebo-controlled study with randomised testing over a 12-week period. Randomisation was done between a moisturiser not containing a niacinamide concentration and one with a 5% concentration. The results showed that the 5% topical solution yielded more noticeable effects than the cream without any niacinamide.
A clinical test was conducted by the Department of Life Science at the University of Seoul. The study found that cells which were exposed to vitamin B3 endured less oxidative damage, results that are consistent with an antioxidant. Studies have also shown that niacinamide may be useful in increasing the elasticity of the skin as well as in the treatment of wrinkles.

Higher concentrations of Vitamin B3 have been shown to lighten skin and so to even out blotch patches. Extended exposure to sun light is a main reason for hyperpigmentation. Melanocytes in deeper skin layers produce melanosomes that contain the pigment melanin. These are then released to keratinocytes that move upwards to the upper epidermis. Niacinamide does not inhibit the production of melanin but Hakozaki et al. showed niacinamide inhibits the transfer of the melanosomes to the surrounding keratinocytes up to 68%. The scientists used a keratinocyte/melanocyte co-culture model. A clinical trial with volunteers confirmed the skin lightening activity. They had applied a skin cream with 5% niacinamide for 8 weeks. Age spots around the eye and cheek were significantly reduced.
